I'm not gonna say Nikon or Cannon, because they are givens. Really a camera comes down to the lens, and how good it is. All the cameras nowdays are about the same. i shoot with a pentax kd20, and i wouldnt change it for the world. Also, depends what your using if for and how big you want the final print :)
I'd say second what SfPJ said..... rent or borrow stuff to try and then purchase. I personally went to Nikon after a friend at work showed me his camera and lent it to me for a few days to play with and I was happy with it so I got one and now I've upgraded...... So try before you buy and talk to other photographers to hear what they have to say...... good luck either way....
there isnt really a single best camera to use per se. if money is no object then go hasselblad w/ leaf digital back for studio work($60k+). sports or nature photographers have had good luck with the Canon 1D w/800L. ($10k+) landscape photographers have used everything from large format field cameras to point and shoots with great result. so it all really boil down to the photographer's skills, and talent.

digital vs film:
im all for digital because it cuts down on the amount of work thats needed before it can be presented. its much more cost efficient as well.

unedited or photoshop:
i personally feel that even a talented photographer can benefit from some post processing work be it in a darkroom (film), or photoshop (digital). some purists will beg to differ though.
